So I have Windows 10, i have converted my user to a Local User so I could use this application.

When i type my password it then says welcome, the YubiKey then begins to flash, so i press it and it says something about Handle error.
Then it says Welcome for about 2 mins then my PC turns off and reboots...

Any ideas?

EDIT: First time I try to logon it says "Remote Procedure Call Failed" then the others give the Handle Error, then my PC restarts.

"Require user input (button press)" should be disabled when you program the Challenge-Response credential, per the instructions. Windows will not give you ample time to respond with button press if you have this option enabled. It will end up failing 19 times out of 20. Boot to Safe Mode, log into your account, run YubiKey Logon Administration to disable the requirement, then follow the instructions again for programming the Challenge-Response credential and return to YubiKey Logon Administration and "Configure" again.
