Hi!
I have installed Windows 2016 Server and Windows 10 on an Oracle VM.
While logging-in User on Windows 10 , currently I can use "either" Password "or" Smart card option (i.e,Yubikey).
Well in this case ,if someone anyhow get the Password then there's no use of Yubikey.

So what i need is the way that User must provide both , Password AND Yubikey to login.

Please advice on how to implement that and if document available?
Appreciate help in this regard.

No, not natively. You either get password only, or smart card + PIN. You add additional requirements you would need a third party integration with something like Duo Security or Okta (many other options listed here - https://www.yubico.com/solutions/#authentication).

Anything more than requiring a smart card plus the PIN is overkill.
