This is covered in Enabling YubiKey with LastPass. The process is the same as for adding the first YubiKey, just enter the backup into the YubiKey #2 field in your LastPass settings. If you're also using both NEOs on Android, you will want to ensure that Offline access is disabled.

OK, so I have a Neo, which I use with LastPass, and just bought another one (as a backup). What do I do now?

That is, how can I link my second Neo to my first Neo, so I can use either?
